Lecture Title: 'Probing basic cellular functions by abiotic stress'

Ramón Serrano is born at Valencia, Spain. He is Doctor Agricultural Enginneer, Research Professor of the spanish CSIC (Research Council) and Full Professor of Biochemistry and Molecular Biology at the Polytechnic University of Valencia, Spain.

Condecorated with the "Cruz de Alfonso X El Sabio" of the spanish government and Doctor Honoris Causa by the University of Regensburg, Germany.
The scientific background of Ramon Serrano is based on the training in the laboratories of three master scientists of modern biology: the spanish enzymologist Alberto Sols, the american bioenergeticist Efraim Racker and the american molecular biologists Gerald R. Fink. Their influence generated an interest at the molecular level on the connection between membrane transport, enzyme activity and cell growth.
The first achievement of Ramon Serrano as independent scientist was the characterization of the proton pump of yeast and plants. In animal cells, Jens C. Skou (Nobel Prize of Chemistry in 1997) discovered in the late fifties an ATPase which coupled ATP hydrolysis to Na and K transport across the plasma membrane. However, this enzyme does not exist in fungi and plants and the bioenergetics of the plasma membrane of non-animal cells was plunged into confusion for many years. Thanks to the work of Ramon Serrano in Spain, together with that of André Goffeau in Belgium and of Carolyn W. Slayman in USA, it became clear that the plasma membrane of fungi and plants is energized by a proton-pumping ATPase, not by a Na-K dependent enzyme.
Ramon Serrano was also at the frontline in the molecular cloning of the gene of the yeast proton pump, work performed in collaboration with Gerald R. Fink at the Whitehead Institute (MIT, Cambridge, Massachussetts, USA). Also, the plant proton pump was cloned for the first time in his laboratory at Heidelberg at the same time than in the laboratory of M.R. Sussman at Madison (USA). These non-animal H-ATPases belonged to the same protein family than the Na-K ATPase. Apparently, in the course of evolution, a primaeval pump had diverged into the animal and non-animal lineages. Animal cells have a primary sodium pump to regulate its volume and avoid swelling. On the other hand, fungal and plant cells obviate this problem by the presence of a rigid cell wall and exploit proton pumping.
Further work of Ramon Serrano defined the essential residues of the proton pump and identified a regulatory domain at its carboxyl terminus as well as a phosphatase domain essential to complete the catalytic cycle by hydrolysis of the phosphorylated intermediate. From the physiological point of view, he demonstrated a connection between cell growth and proton pump activity in yeast. Active nutrient transport and pH regulation were implicated in this important phenomenon. In order to extrapolate these results to animal cells, the yeast proton pump was expressed in fibroblast, which experienced elevated intracellular pH and increased proliferation (in collaboration with Rosario Perona, Madrid). In plants, the effect of auxin on cell growth was correlated with induced expression of the proton pump (in collaboration with Achim Hager, Tubingen).
Since 1990, Ramon Serrano started to work on the molecular basis of proton, potassium and sodium homeostasis, utilizing the yeast Saccharomyces cerevisiae and the plant Arabidopsis thaliana as model system. This was another aspect of the connection between membrane transport and cell growth and had the social relevance that it could generate the biotechnological tools to improve the salt tolerance of crops, an urgent need for the future of agriculture in irrigated land. This work identified the first target of sodium toxicity in living cells, the nucleotidase encoded by the yeast HAL2 gene. This enzyme is very sensitive to inhibition by Li and Na and it is required to detoxify a metabolite, adenosine-3',5'-diphosphate or PAP, which inhibits sulfate assimilation, sulfate transfer and RNA processing. Plant HAL2 homologues were characterized which could be targets of sodium toxicity. The Hal2 protein has been crystallized and the molecular basis of Li and Na inhibition characterized at the atomic level (in collaboration with Armando Albert at Madrid and Tom Blundel at Cambridge).
The transcription factor Sko1 has been identified by his laboratory as a repressor of some defense genes modulated by the yeast MAP kinase Hog1, which is activated by salt stress. This established a novel paradigm of negative control for salt stress defense genes such as ENA1 (encoding a Na+ extrussion pump) and HAL1 (encoding an activator of K+ transport). The identification of protein kinase (HAL4 and HAL5) and protein phosphatase (PPZ1) genes as modulators of K+ transport opened the molecular bases of ion homeostasis in yeast. Ppz1 is inhibited by Hal3 and the Hal3-Ppz1 interaction is pH-dependent, constituing the first pH sensors identified in cells.
Latest works of Ramón Serrano have established a connection between plant sulphydryl oxidases and ion homeostasis through regulation of potassium transport at the xylem loading interface. Also, oxidative stress has been found to desensitize ethylene responses in Arabidopsis.

Lecture Title: 'Plant-Insect Interactions: the Potential Role of Cross-Talk in
Caterpillar-Mediated Circumvention of Plant Defense Responses'

Research:
Plants adjust their defense responses to target herbivores. In response, insect herbivores have evolved adaptive strategies to undermine these plant defenses. Research in Bede's laboratory lies at the boundary of plant-caterpillar interactions, characterizing the co-adaptive responses between plant and caterpillar herbivores at the molecular, biochemical and physiological levels. Understanding the underpinnings of these complex interactions is critical to gain insight into the complex molecular evolution between plants and insects and develop effective pest management strategies.
